Profilin is a protozoan parasite protein. It activates toll-like receptor 11 (TLR11), which is an intracellular receptor localized on the endoplasmic reticulum (ER). TLR11 acts as an innate immune sensor for T.gondii profilin (TgPRF).The 12 membrane-spanning ER-resident protein UNC93B1 directly binds TLR11 and control the activation of dendritic cells in response to profilin (Toxoplasma gondii) and parasitic infection in vivo. TLR12 also has an ability to interact with (TgPRF). TLR12 is adequate for recognizing profilin in plasmacytoid dendritic cells (pDCs), whereas TLR11 and TLR12 are both needed in macrophages and conventional DCs. In contrast to TLR11, TLR12-deficient mice fail to resist T. gondii infection.
