Biochem/physiol Actions

EIF4E (eukaryotic translation initiation factor 4E) is a part of eIF4F complex, which is responsible for the recruitment of ribosomes to the capped end of mRNAs during mRNA translation. It acts as the rate-limiting element in cap-dependent mRNA translation in eukaryotic cells. It promotes the translation of pro-oncogenic mRNAs, including vascular endothelial growth factor (VEGF), cyclin D3, and Mcl-1 (induced myeloid leukemia cell differentiation). Up-regulation of this protein is linked with various cancers, such as prostate cancer, breast cancer, and lymphoma. Studies show that this protein is overexpressed in melanoma cell lines and is linked with resistance to vemurafenib. Thus, it might have potential as a therapeutic target to suppress melanoma cell proliferation and abrogating vemurafenib resistance.

General description

EIF4E (eukaryotic translation initiation factor 4E) is a m7GpppN (N is any nucleotide) cap-dependent RNA-binding protein. It is one of the three subunits of the translation initiation factor complex eIF4F, and is the least common of all initiation factors.

Physical form

Formulated in 50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 0.25 mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF, 50% glycerol.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
