Biochem/physiol Actions

HDAC6 (histone deacetylase 6) functions as a microtubule or tubulin deacetylase, which in turn regulates microtubule-dependent chemotactic cell motility. This protein shows colocalization with p150glued protein, a dynactin component. It contains a ubiquitin binding zinc finger, and interacts with ubiquitinated proteins post-proteasomal inhibition. It is essential for the survival of inflammatory breast cancers (IBCs), though does not function as an oncogene. Treatment of IBC cells with HDAC6 inhibitor ricolinostat (ACY1215) results in the inhibition of IBC cell proliferation both in vitro and in vivo.

General description

HDAC6 (histone deacetylase 6) is a member of the HDAC family of enzyme which are generally linked with gene expression and chromatin dynamics. This protein resides exclusively in the cytoplasm and is present in the perinuclear punctate structures as well as the leading edges of the cell.

Physical form

Formulated in 40 mM Tris-HCl pH8.0, 110mM NaCl, 2.2 mM KCl, 0.04% Tween20, 3mM DTT, and 20% glycerol.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
