Application

Kinase activity is measured as the molar amount of phosphate incorporated into the CREBtide substrate peptide per minute per mg protein at 30 °C using a final concentration of 50 µM [32P] ATP.

Biochem/physiol Actions

cAMP-dependent protein kinase (PKA; EC 2.7.1.37) is an essential enzyme in the signaling pathway of the second messenger cAMP. Through phosphorylation of target proteins, PKA controls many biochemical events in the cell including regulation of metabolism, ion transport, and gene transcription.

General description

Protein kinase cAMP-activated catalytic subunit ? (PRKACB) is one of the catalytic subunits of the serine/threonine kinase, protein kinase A. The gene encoding it is localized on human chromosome 1p36.1.

Physical form

Supplied as a solution of 5 µg in 50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 0.25 mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF and 25% glycerol.

Unit Definition

One unit will incorporate one nanomole of phosphate into the Crebtide (KRREILSRRPSYR) substrate per minute at 30 deg C and pH 7.2 using a final concentration of 50 µM [32P] ATP.
