The denaturing solutions is composed of 0.5N NaOH, and 1.5M NaCl.
General description
Sigma’;s denaturing solution is a strong base that denatures double-stranded DNA so that it can be efficiently transferred to nylon membrane and subsequently hybridized to labeled probe. When used with a depurination solution, treatment results in the hydrolysis of the phosphodiester backbone at the sites of depurination, allowing for more efficient transfer of large DNA in neutral Southern transfers.
