General description


Transducin-like enhancer protein 1 (UniProt Q04724; also known as E(Sp1) homolog, Enhancer of split groucho-like protein 1, ESG1) is encoded by the TLE1 gene (Gene ID 7088) in human. TLE proteins interact with HDACs as well as deacetylated histone tails and play an important role in transcription regulation in several signaling pathways, including Ras, Notch, and Wnt. TLE proteins contain a conserved N-terminal glutamine-rich (Q) domain (a.a. 1-131 of TLE1) that mediates TLEs tetramerization as well as TCF/LEF proteins binding, and is essential for chromatin oligomerization. This is followed by a variable central domain sub-divided into a glycine-prolin-rich (GP) region (a.a. 132-199 of TLE1), the CcN (protein kinase CK2/cell cycle-dependent kinase 2/nuclear localization sequence) domain (a.a. 200-268), a serine-proline-rich (SP) region (a.a. 200-268 and 269-450 of TLE1), and a C-terminal WD40 domain with 6 WD repeats (a.a. 470-767 of TLE1) that interact with multiple transcription factors. The GP region binds directly to HDAC1, the CcN region contains consensus CK2 and Cdc2 phosphorylation sites that are modified in a cell cycle-dependent manner, the SP region is phosphorylated by the Ser/Thr kinases HIPK2 and MAPK, which appear to negatively regulate transcription repression activity, and may also interact with basic helix-loop-helix transcription factors. The C-terminal WD40 domain is essential for chromatin condensation and, together with SP, interacts with bHLH proteins.
